First participant dosed in Tangram’s TGM-148 trial at Nucleus Network London

Nucleus Network London is proud to support Tangram Therapeutics’ Phase 1 trial of TGM-148, an investigational RNAi medicine being developed for bleeding disorders.
Tangram announced the dosing of the first participant on 29 October 2026. TGM-148 is the company’s second GalOmic RNAi medicine to enter the clinic in 2026, marking another milestone for its development pipeline.
The program’s planned first indication is von Willebrand disease (VWD), the most common inherited bleeding disorder. People living with severe VWD can experience frequent bleeding and significantly reduced quality of life. Preventive treatment options remain limited and burdensome, potentially involving multiple intravenous infusions each week.
TGM-148 is designed to selectively silence Protein Z-dependent Protease Inhibitor (ZPI), a natural regulator of coagulation, in the liver. Its mechanism aims to enhance clot formation at sites of active bleeding when clotting is impaired, while maintaining a favourable safety profile.
With an anticipated regimen of four or fewer subcutaneous doses a year, Tangram aims to develop a preventive treatment that could reduce the burden of prophylaxis and fit more easily around patients’ lives.
The Phase 1 trial will evaluate the safety, tolerability, pharmacokinetics and pharmacodynamics of single ascending doses in healthy volunteers. Initial data are anticipated in 2027 and will help inform progression into patient studies.
Preclinical studies demonstrated efficacy in four genetic models of bleeding disorders. These findings support the rationale for investigating TGM-148 in VWD, with the potential to explore other rare bleeding disorders as development progresses.
We congratulate Tangram on this milestone and thank the participants and study teams at Nucleus Network London who make this research possible.
